The transgene comprises a 0.41-kb DNA fragment containing the rat insulin 1 promoter, a 1.6-kb mouse cDNA encoding an in-vitro mutated variant of fyn-related kinase (FYN) joined to a cDNA fragment encoding a MYC epitope tag, and the polyadenylation signal of the bovine growth hormone gene. The mutant fyn-related kinase protein has a substitution of phenylalanine for tyrosine at amino acid position 504, which confers enhanced kinase activity. Expression of transgene-derived mRNA was demonstrated by polymerase chain reaction of islet cDNA using a primer matching the Myc sequence. Only moderate elevation of FYN protein was detected in islet lysates or in FYN immunoprecipitates using unpurified anti-FYN antiserum. (J:93623)
