A DNA segment encoding a truncated version of bone morphogenic protein receptor, type II (serine/threonine kinase), preceded by an internal ribosome entry site (IRES) and followed by a frt-flanked neomycin resistance cassette, was introduced into the 3' untranslated region of the endogenous tyrosine hydroxylase gene. In situ hybridization and immunohistochemcal analysis revealed reduction in the levels of tyrosine hydroxylase mRNA and protein in brains of homozygous mice. (J:93552)
