这个等位基因利用了含Hoxd11/lac/loxP报告基因的胚胎干细胞系，该基因位于Hoxd13的5'端。在Hoxd1基因的3'端插入了反向定向的floxed neo。与Cre在生殖细胞中表达的鼠杂交，会剔除neo并反转较大段的floxed序列（来源：J:91567）。
