The two coinjected transgenes comprise functionally rearranged genomic T-cell receptor alpha and T-cell receptor beta genes that recognize the male specific H-Y antigen in the context of the major histocompatibility (H2) class II I-Ab29 protein. The joined V-J segments of the T-cell receptor alpha transgene and V-D-J segments of the T-cell receptor beta transgene were isolated from the CD4 T-cell clone "Marilyn" and inserted into expression, or shuttle, vectors that contain genomic sequences including the constant-region exons of Tcra and Tcrb and their associated promoters and enhancers.
