在细胞中，通过同源重组技术，将包含带有floxed tk-neo组件的tau-GFPpA报告基因 cassette替换掉了前5个外显子，仍由内源性启动子调控转录。编码的前162个氨基酸区域包含了对DNA结合和蛋白结合位点转录激活至关重要的区域。随后的Cre介导重组移除了tk-neo cassette。原位杂交结果显示没有检测到内源基因的表达。GFP的表达模式与内源基因一致。（来源：J:88585）
