在 intron 2 中插入了floxed的诺卡因抗性基因，同时在exon 1中插入了loxP位点。构建出的产物是一个表达量减少80%的弱变种，蛋白质产物也相应减少。通过将针对的ES细胞短暂转染一个cre表达载体，以去除诺卡因但保留exon 1和2，且这两个区域被loxP位点环绕，也得到了一个表现出相似表型的弱变种。（来源：J:86857）
