通过同源重组，PGK-neo cassette被插入到了exon 1内。neo序列位于信号肽下游，导致了14个核苷酸（268-282）的糖蛋白区段被删除，并产生了框架突变。对纯合突变小鼠的RT-PCR分析显示，存在一个缺失了5'端编码信号肽和大部分糖蛋白区段的框架移位转录本。尽管这个突变转录本包含了77个neo cassette序列、exon 1的3'端以及exons 2到11，但胃组织的 Western blot检测并未发现蛋白质。(来源：J:80329)
