内源性位点的Exon 2被一个包含β-gal和neo基因的 cassette 替换，产生了特异性的等位基因变异。Exon 2是Citron-K亚型的第二个编码外显子，位于Citron-N转录起始点的上游。在杂合突变小鼠的脑、脾和胸腺组织的 Western blot 检测中，未检测到Citron-K亚型，而Citron-N亚型的水平正常。（来源：J:77297）
