CD4的内源性调控元件核心区域，在T细胞发育期间抑制其表达（S4）的区域，通过缺失167到257号核苷酸的靶向构建的同源重组被删除于第1 intron。保留了一个loxP位点在删除区域上游，但对照等位基因分析表明，这个loxP位点不影响正常表达。对突变小鼠获取的外周血淋巴细胞进行流式细胞术分析，并未发现CD4表达的沉默现象。（J:80576）
