LoxP sites flanking the coding region of the gene were introduced into exon 2 and a floxed tk-neo cassette and lacZ gene were inserted into the downstream region via homologous recombination. Northern blot analysis confirmed expression of the mutant allele, and Western blot analysis showed protein expression is similar in mutant animals compared to wild-type. (J:78662)
