A floxed region containing neo and a transcriptional blocking sequence (STOP) was followed by human sequence encoding a modified SNCA gene. This construct was incorporated 7 bp upstream of the endogenous start codon via homologous recombination. Northern and Western blot analyses of homozygous mutant mice indicated a complete ablation of gene expression. The positioning of the human sequence downstream of the transcription blocking cassette precluded its expression. (J:79993)
