通过将Cre重组酶/改良的雌激素受体(ESR1)融合序列插入内源性等位基因，产生了一个特异于组织且依赖于配体的重组酶。ESR1序列在400、543和540位点进行了工程性突变，这些突变替换掉了与内源性配体结合的能力，转而能与他莫昔芬结合(参考文献J:79835)。对接受他莫昔芬处理的突变小鼠进行组织化学分析，结果显示Cre在平滑肌组织中表达(参考文献J:69325)。
