An intronic floxed neo cassette was removed by crossing to a cre-expressing deleter strain, leaving behind a tyrosine to phenylalanine knock-in mutation at position 315 (Y315F). Expression of the mutant allele was confirmed by sequence analysis of RT-PCR products from thymus of mutant animals. (J:71099)
