The gene was disrupted by replacement of a large portion of exon 1 with a PGK-neo cassette. The targeted mutation deletes sequences encoding amino acids 22-150. Northern blot analysis and quantitative RT-PCR was unable to detect gene transcripts from hepatic mRNA of homozygous mutant animals, thus confirming the null allele. (J:71785)
