这个构建中，编码区的起始部分被一个上游的floxed neo cassette和下游的loxP位点包裹，通过同源重组实现了整合。对脾细胞进行流式细胞术分析后发现，loxP位点和neo cassette的插入并未影响MHC-II表面表达（来源：J:75115）。
