在转录起始密码子上游272bp的位置插入了loxP位点，然后在exon 1内插入了loxP-tauGFP带有FRT标记的neo cassette，通过同源重组方式进入内含子1，位置在1kb处。杂合子动物与flp重组酶转基因品系杂交以去除neo cassette。接着与cre重组酶转基因品系杂交，移除了exon 1和部分内含子1，保持tauGFP cassette完好。exon 1的删除去除了膜靶向信号序列，并使tauGFP cassette受内源性启动子控制。RT-PCR结果显示，纯合突变动物基因转录本中信号序列已被去除。对纯合突变动物大脑切片的免疫组化未检测到蛋白质产品。(来源：J:76686)
