An IRES-lacZ gene followed by a neomycin selection gene and thymidine kinase cassette replaced a genomic fragment containing exons 2-6. RT-PCR analysis on total brain RNA derived from homozygous mice demonstrated that no detectable transcript is produced from this allele. Western blot analysis on lysates of brain tissue of homozygous mice confirmed that no endogenous protein is expressed from this allele. However, brain histology revealed that a beta-galactosidase protein is expressed from this allele under the control of the endogenous promoter. (J:70254)
