Homologous recombination introduced a floxed neomycin resistance cassette into intron 2, and a single loxP site into intron 1. Cre-mediated recombination in vivo under the control of a CMV promoter excised the neo cassette in the germline, leaving single loxP sites flanking exon 2, which encodes roughly a third of the extracellular domain of the receptor within the ligand-binding domain.
