A genomic fragment containing exon 3 was replaced with a neomycin selection cassette in opposite transcriptional orientation to the gene. RT-PCR analysis detected the presence of a trancript that spliced exon 2 to 4, but this mutation introduces a frameshift and premature stop codon. Western blots on testicular extracts confirmed the absence of the full length protein in homozygous mice. (J:58730)
