在exon 5的5'端插入了loxP引导的hygromycin抗性基因座，而在exon 5的3'端则插入了一个单独的loxP位点，编码磷酶活性区。在将hygromycin基因座通过Cre重组酶的暂时表达从胚胎干细胞中去除后，再生成嵌合小鼠，留下的位置是基因座原本的位置。这些插入似乎对基因的正常功能没有影响。（来源：J:75117）
