Insertion of an in-frame lacZ gene and neomycin cassette into the sequences encoding the first exon. A frameshift mutation at an Sst1 site in the sequences encoding the homeobox domain was also made by cutting with the enzyme, blunt ending and ligating back together. No wild-type protein was detected in E9.5 embryos as assayed by whole mount immunohistochemisty. (J:37450)
