将靶向载体插入后，产生了一个包含抗氨苄青霉素抗性基因座的重复exon 1。上游（插入的）exon 1在第12位由甘氨酸突变为天冬氨酸，导致p.G12D变异。在无同染色体重组的情况下，这个等位基因不会产生蛋白质，纯合突变体胚胎期会死亡。杂合子中，体内同染色体重组发生时，一半事件产生编码p.G12D氨基酸替换的活性突变等位基因，另一半产生野生型等位基因。G12D蛋白的表达会导致肺肿瘤形成并在约300天（10个月）时导致死亡。（来源：J:68981）
