一种由诺威霉素抗性基因 cassette（带有loxP位点，可剪切的）替换基因的exon 3。携带有这种突变基因的鼠与从胚胎早期就持续表达Cre重组酶的鼠杂交。产生的携带所需删除突变的鼠相互交配，产生了在目标位点上纯合突变鼠。对纯合突变鼠的胸腺细胞和抗CD3刺激的脾细胞进行Northern blot检测，未检测到目标基因的可检测mRNA。（来源：J:42508）
