经过修改的D3胚胎干细胞系，其Selptm1Hyn等位基因为杂合状态，再次被定向编辑。信号肽编码区域、凝集素域以及表皮生长因子部分区域被插入了PGK-hygro载体。对心脏和肺组织的提取物进行的Northern和RT-PCR结果显示，这两基因在纯合突变小鼠中均无正常转录信息。这个靶向突变与Selptm1Hyn突变发生在同一条染色体上，由于临近的基因组位置，它们应随后代一同遗传。(来源：J:31626)
