A neomycin resistance cassette replaced a 1.3 kb region of the gene, which spanned the translational start codon, the signal peptide coding region, and most of exon 1 of the gene. In the construct designated bb1, the neomycin cassette was inserted in the same transcriptional orientation as the gene; in the construct designated bb2, the neomycin cassette was inserted in the reverse orientation.
