Using CRISPR/Cas9 technology, the mouse Xist gene, driven by human eukaryotic translation initiation factor 1A X-linked (EIF1AX) promoter was inserted downstream of the Runx1 locus on chromosome 16 (a low-gene density region designated as Igs50) of the trisomy chromosome 16 in the host ES cell derived from B6EiC3Sn a/A-Ts(17<16>)65Dn/J line.
