The targeting construct replaced exon 3 with one containing the mutation responsible for the amino acid substitution of leucine to alanine at position 43 (L43A). This mutation abolishes the function of the RAS-Binding domain. Flp-mediated recombination removed the FRT-flanked neomycin resistance cassette. (J:331473)
