The allele was created by first inserting an Actb-neo cassette followed by a loxP (fl) site at position 116,237,220 (mm9, Chr.12) and blasticidin resistance gene (Bsd)-rox sequence into the middle of the VH gene cluster, followed by the insertion of a frt-Pgke promoter sequence-frt and a second lox (lox71) site (in the same orientation) at position 117,126,667 followed by Puro cassette in the Igh 5' region.
