An IRES-CreERT2-IRES-humanCD4-FRT-neo-FRT construct was inserted into the 5th exon of the endogenous Prtn3 gene after the endogenous stop codon. The Neomycin cassette used for selection was then removed by the flippase enzyme. The human CD4 coding sequence used in the construct lacks the C-terminal 33 amino acids and contains the F43I mutation, generating a mutated and truncated protein that precludes signaling and association with MHCII.
