Fertilized oocytes were coinjected with two enhanced green fluorescent protein (EGFP) transgene constructs. One construct, comprising the chicken beta-actin promoter and the cytomegalovirus immediate early enhancer followed by the EGFP coding sequence and the rabbit beta-globin polyadenylation signal, directs expression of EGFP to the cytoplasm of most cell types; sperm of mice having only this construct contain too little cytoplasm to fluoresce. The second construct, containing the mouse preproacrosin promoter and sequences encoding the acrosin signal peptide and N-terminal peptide fused to the EGFP coding sequence, directs EGFP expression specifically to the sperm acrosome, which therefore flouresces.
