The targeting construct contains the Pax7 open reading frame (ORF), a Kozak sequence to allow protein translation initiation. An FRT-flanked puromycin cassette inserted downstream of exon 8 followed by exon 9, a T2A self-cleaving peptide, ZsGreen1 fluorescent reporter sequence, and 3 nuclear localization signals (3xNLS) inserted upstream of the 3' untranslated region (UTR), Flp-mediated recombination removed the puromyxin cassette.
