A construct containing a Pgk promoter driving a tetracycline repressor gene followed by a poly a sequence and a tetracycline responsive Pol III hybrid promoter driving expression of Pum3 shRNAs. Treatment with doxycycline results in about an 80% reduction in Pum3 mRNA expression in hemizygous embryos. (J:338852)
