The human tau gene exons 2-12 (from BAC RP11-111I23) flanked with loxP sites replaced the mouse genomic sequence from the translation initiation codon in exon 2 to the termination codon in exon 10. The gene Saitohin (STH) is present in the human tau gene and is included in the targeting vector. In addition, an FRT-flanked neomycin resistance cassette was inserted downstream of the first loxP site and was removed via flp-mediated recombination. Murine and human tau-specific antibodies confirmed the absence of the mouse protein and presence of the human protein, respectively. (J:338832)
