This transgene consists of a 0.9 kb DNA fragment containing the cytomegalovirus early gene promoter fused to the rabbit beta-globin second intron, a loxP flanked chloramphenicol acetyltransferase (cat) cassette connected to a polyadenylation signal, followed by a 1.4 kb DNA fragment encoding a bovine dominant-negative mutant for Rho-associated coiled-coil containing protein kinase 2, which contains two point mutations to suppress Rho-binding activity, and a second polyadenylation signal. Upon cre-mediated excision of the cat gene, the modified allele expresses the mutant ROCK2 in tissues where cre is expressed. Two lines were generated, 3-1, 3-3, 4-9, 4-11, 5-18, and 5-27.
