The following were inserted into intron 1 of the locus: splice acceptor sequence, a loxP site flanked neomycin resistance gene cassette and STOP cassette, a modified human AKT1 cDNA (where a G-to-A point mutation changed glutamic acid codon 17 to lysine (p.E17K) and codon-optimized for mouse expression), IRES sequence, the EGFP reporter gene, and poly(A) signal sequences. The inserted sequence is only expressed after Cre-mediated deletion of the neo-STOP cassette. (J:307913)
