A loxP site was inserted upstream of exon 3 and a second loxP site, FRT-flanked neomycin resistance cassette, and third loxp site in opposite orientation were inserted downstream of exon 3. Flp-mediated recombination removed the neomycin resistance cassette leaving exon 3 floxed. (J:246331)
