The transgene containing the murine Camk2a promoter drives expression of the tamoxifen-inducible cre recombinase gene (cre/ERT2) fused with the oxygen-dependent degradation (ODD) domain of Hif1a. This transgene is responsive to hypoxia. Under normoxic conditions, the ODD is hydroxylated by prolylhydroxylases, which tag HIF-1 upon binding of Von Hippel-Lindau (VHL) protein for proteasomal degradation. Hypoxic conditions lead to the inactivation of these enzymes and thus stabilization of HIF-1, allowing it to accumulate in the nucleus and activate the transcription of its target genes.
