The targeting construct containing IRES-FlpO-a LoxP-flanked neomycin selection cassette was targeted 3 bp downstream of the stop codon of the Slc17a6 gene in ES cells. Targeted ES clones were expanded and injected into blastocysts and chimeric offspring were then bred to confirm germline transmission of the allele. The neomycin selection cassette was removed by breeding to the E2A-Cre deleter strain. (J:317381)
