The transgene insert contains the following elements: the 5' PiggyBAC transposon inverted repeat, 1027 bp sequence containing Atoh1 enhancer Eh2 or E3, Hsp68 minimal promoter, EGFP reporter gene, SV40 early poly(A) signal sequences, and the 3' transposon inverted repeat. The transgene was injected into single-cell zygotes together with PiggyBAC transposase mRNA to facilitate integration into the genome. The reporter is expressed in cochlear hair cells in newborn mice. (J:333396)
