The targeting vector is constructed based on the Ai6 vector from Addgene with (from 5' to 3') a CAG (CMV enhancer/chicken beta-actin) promoter, an FRT site, a floxed STOP cassette (with stop codons in all 3 reading frames and a triple polyA signal), and murine B cell leukemia/lymphoma 2 (Bcl2) cDNA followed by a woodchuck hepatitis virus post-transcriptional regulatory element (WPRE), a polyA signal, and an attB/attP-flanked PGK-FRT-Neo-polyA cassette. The targeting vector is inserted into the Gt(ROSA)26Sor locus. (J:334079)
