This transgene was created by coinjection of two overlapping genomic fragments: an NcoI-NcoI restriction fragment containing sequence 5' to and 3' from the Atoh1 coding sequence and a BglII-BglII fragment containing the last ~400 bp of coding sequence and 6 kb 3' of the coding region. Coding sequence from the N terminus of Drosophila Bride of Sevenless (BOSS) (TSPTKKSAPLRITKPQPTS) was inserted in-frame with the Atoh1 coding sequence after codon 8 to enable distinguishing endogenous from transgenic gene products.
