The targeting vector was designed with a CAG promoter, an FRT site, a loxP-flanked STOP cassette, a Flag-CaMPARI (calcium-modulated photoactivatable ratiometric integrator) sequence, a woodchuck hepatitis virus post-transcriptional regulatory element (WPRE; to enhance the mRNA transcript stability), a poly-A signal, and an attB/att-flanked PGK-FRT-Neo-poly-A cassette. This entire construct was inserted into the Gt(ROSA)26Sor locus via electroporation of C57BL/6-derived embryonic stem (ES) cells (J:267056)
