The transgene comprises a 129 strain-derived (Igha haplotype) BAC, containing an anti-arsonate (ARS) re-arranged VDJ region plus all S regions and C segments and the 3'RR super-enhancer (Rr152), that was modified as follows: two copies of chicken beta-globin genomic insulator sequence were inserted upstream of the Igh cluster, 4-bp sequences were inserted in the Ighg1 and Ighg2a genes (to distinguish transgene transcripts from endogenous transcripts), FLAG tag encoding sequence was inserted at the 3' end of the secreted isoform of Ighg2a, and loxP sites were inserted up- and downstream of Rr152.
