A reversed flipflox vector, consisting of a gene inactivation cassette (an internal ribosome entry site (IRES)-GFP reporter in reverse orientation) flanked by mutant loxP sites and an FRT-flanked PGK-neomycin selection cassette, was inserted into intron 3. The neomycin selection cassette was removed via flp-mediated recombination. The gene-inactivation cassette is transcriptionally silent and does not affect TFPI expression. Cre-mediated recombination is necessary to permanently invert the gene inactivation cassette, express EGFP and block transcription of Tfpi. (J:288197)
