The targeting vector was designed to replace the endogenous STOP codon of the gene with a SMASh cassette with a STOP codon, and a loxP-flanked neomycin (neo) cassette. The SMASh (Small Molecule-Assisted Shutoff) cassette contains a cis-cleaving hepatitis C virus (HCV) NS3 protease. Cre-mediated recombination removed the neo cassette. (J:101977)
