Exons 2-4 were replaced by a cassette consisting of a transcriptional repressor sequence (STOP) flanked by loxP sites, cDNA sequence for the first three coding exons (2-4) with 93 rare codons converted to the most commonly used codons in the mouse genome and including the p.Q61R oncogenic mutation, followed by the 5' 564 bp of intron 4, and an FRT site flanked neomycin resistance gene cassette. The neo cassette was removed via Flp-mediated recombination. Removal of the stop cassette is required to get expression of the mutant Kras. (J:328899)
