An FRT site flanked neomycin resistance gene cassette and a loxP site were inserted upstream of the Hnf1aos1 gene promoter/1st exon and a second loxP site downstream of the Hnf1aos2 gene promoter/1st exon. These exons, promoters and loxP sites are located in Hnf1a intron 1. The neo cassette was removed through subsequent flp-mediated recombination. The ~1.8 kb floxed sequence, containing Hnf1aos1 and Hnf1aos2 promoters and 1st exons, was subsequently deleted through cre-mediated recombination. (J:332319)
