The transgene insert contains the following elements: an FRT site flanked neomycin resistance gene cassette, Gata2 upstream regulatory sequence (from ~4 kb upstream of exon 1S to ATG translation start in exon 2; contains upstream alternative promoter, 1st exon, and intron 1S and downstream alternative promoter, 1st exon and intron 1G, and 5' UTR part of exon 2), the lacZ reporter gene, and BAC RP23-333I12 (which contains the 5' end of Eefsec with Gata2 enhancers Rr341 and Rr342 in its introns). The neo cassette was removed through subsequent flp-mediated recombination. The reporter gene is expressed in urogenital mesenchymal (but not epithelial) tissue, and digits, boundary cap cells, otic vesicle, hind- and midbrain, nose and other tissues. (J:95123)
