The targeting vector is designed to replace the stop codon in exon 18 with a bicistronic reporter containing a P2A-H2B-Venus non-gene disruptive reporter cassette. The H2B-Venus nuclear reporter is a fusion protein consisting of the coding sequence of human histone 2B (human HIST1H2BB gene [histone 1 H2bb]) N-terminally fused to the yellow fluorescent protein Venus followed by an FRT-flanked neomycin selection cassette. Flp-mediated recombination removed the neo cassette. (J:101977)
