A loxp flanked TK/neomycin selection cassette was inserted upstream of exon 2 and a third loxP site was inserted downstream of the exon 2 coding region within the non-coding region of the exon. The selection cassette was removed via cre-recombinase transfection in ES cells leaving exon 2 floxed. (J:269358)
